{Abstract}: A water-soluble glucan (PS-I) was isolated from the aqueous extract of the fruit bodies of a hybrid mushroom, pfls 1h of Pleurotus florida and Lentinus squarrosulus (Mont.) Singer. Structural characterization of PS-I was carried out using total hydrolysis, methylation analysis, periodate oxidation, and NMR experiments (H-1, C-13, DEPT-135, DQF-COSY, TOCSY, NOESY, ROESY, HSQC, and HMBC). Methylation analysis revealed that PS-I was composed of (1 -> 3, 6), (1 -> 3), (1 -> 6)-linked and terminal beta-D-glucopyranosyl residues in a relative proportion of approximately 1:1:1:1. The repeating unit of the glucan consists of a backbone chain of two (1 -> 6)-beta-D-glucopyranosyl residues, one of which is branched at O-3 position with (1 -> 3)-beta-D-glucopyranosyl and terminated with a beta-D-glucopyranosyl residue. Study of immunological activity revealed that PS-I stimulates the splenocytes, thymocytes and macrophages. (c) 2012 Elsevier B.V. All rights reserved.

{Abstract}: A heteroglycan (PS) isolated from the mycelia of Pleurotus ostreatus was found to consist of L-fucose, D-mannose, and D-glucose in a molar ratio of nearly 1:2:3. On the basis of acid hydrolysis, methylation analysis, periodate oxidation, and NMR studies (H-1, C-13, TOCSY, DQF-COSY, NOESY, ROESY, HSQC, HMBC, and DEPT), the structure of the repeating unit of the PS was determined. The repeating unit had a branched backbone composed of (1 -> 6)-linked alpha and beta D-glucose and (1 -> 2)-linked alpha-L-fucose. Branching occurred at C-4 position of (1 -> 6)-linked alpha-D-glucopyranosyl residue with terminal beta-D-mannose and C-3 position of (1 -> 6)-linked beta-D-glucopyranosyl residue with (1 -> 6)-alpha-D-mannopyranosyl moiety terminated by alpha-D-glucose. The structure of the repeating unit of the PS was proposed as:

The antioxidant properties of the PS were studied. The scavenging activity of hydroxyl free radicals whose EC50 value of PS at 943 mu g/mL was determined. Further, EC50 value of the PS at 53 mu g/mL was observed for scavenging activity of superoxide free radicals. Chelating effects (54.82%) of ferrous ions were observed at 1 mg/mL of the PS. {Abstract}: The optimal culture conditions of exopolysaccharides (EPS) production in submerged culture medium by Pleurotus geesteranus 5 # were determined using an orthogonal matrix method. The optimal defined medium (per liter) was 60.0 g maltose, 5.0 g tryptone, 1 mM NaCl, 5 mM KH2PO4, and initial pH 6.0 at 28 degrees C. In the optimal culture medium, the maximum EPS production was 16.97 g/L in a shake flask. Two groups of EPSs (designated as Fr-I and Fr-II) were obtained from the culture filtrates by size exclusion chromatography (SEC), and their molecular characteristics were examined by a multiangle laser-light scattering (MALLS) and refractive index (RI) detector system. The approximate weight-average molar masses of the Fr-I and Fr-II of EPS were determined to be 3.263 x 104 and 5.738 x 103 g/mol, respectively. The low values of polydispersity ratio (1.176 and 1.124 for Fr-I and Fr-II, respectively) of EPSs mean that these EPS molecules exist much less dispersed in aqueous solution without forming large aggregates. Furthermore, the experiments in vitro indicated that P. geesteranus 5# EPS exhibit high antitumor and antioxidative effects.

{Abstract}: A novel water-soluble polysaccharide named PGPB-1, with a molecular weight of 1.3 x 10(4) Da as determined by high-performance liquid chromatography (HPLC), was isolated from the fruiting bodies of Pleurotus geesteranus by boiling water as well as DEAE-Sepharose Fast Flow and High-Resolution Sephacryl S-300. On the bases of compositional analysis, methylation analysis and NMR spectroscopy (H-1 NMR, C-13 NMR, 2D-COSY, TOCSY, HMQC, HMBC and NOESY spectra) analysis, the structural elucidation of PGPB-1 consisted of a alpha-D-(1 -> 6)-galactopyranan and a alpha-D-3-O-Me-D-galactosyl unit backbone with a alpha-D-Mannosyl unit on O-2 of the 2.6-di-O-substituted-D-galactosyl units. The repeating unit of the polysaccharide was established as:
