{Abstract}: Antioxidant properties of exopolysaccharides (EPS) and intracellular polysaccharides (IPS) obtained respectively from mycelia and filtrates of submerged culture by Armillaria mellea in a 20-L stirred tank bioreactor were investigated. Effective production of EPS and IPS is available by submerged culture of A. mellea with respective number average molecular weights and protein/polysaccharide ratios as 7.68x10(6) Da and 7.68% as well as 5.65x10(6) Da and 5.26%. Both EPS and IPS exhibit powerful antioxidant activities by conjugated diene method, chelating effect on ferrous ions and scavenging effect on superoxide anion, as evidenced by their quite low EC(50) values (< 5 mg/mL). Results confirmed that fermented A. mellea polysaccharides are potential antioxidant sources of both healthy medicine and food industries.

{Abstract}: Cordyceps jiangxiensis, also called 'CaoMuWang', is a medicinal entomopathogenic macrofungus native to eastern China. Polysaccharide fractions from cultured C. jiangxiensis exhibited potent antitumor activity via the induction of cell cycle arrest and apoptotic pathway. Antioxidant pathway is also one action of mechanism of antitumor; thus, the antioxidant abilities of these polysaccharide fractions were overall evaluated by five in vitro assays such as the scavenging abilities on DPPH center dot, hydroxyl and superoxide anion radicals, the reducing power and the chelating ability on ferrous ions. Among these assays, the polysaccharide fractions presented more excellent scavenging abilities on superoxide anion radicals than that of the positive control. When compared with the positive control, the polysaccharide fractions from C. jiangxiensis only had moderate scavenging activities on both DPPH and hydroxyl free radicals, moderate reducing power and ferrous ion chelating activity. The antioxidant abilities of the different polysaccharide fractions had certain differences at all the tested doses and all had a dose-dependent manner. The results suggested that, polysaccharides are important antioxidant component in the medicinal Cordyceps fungi and have direct and potent antioxidant ability, and that C. jiangxiensis also is a promising potential source for the development of natural antioxidant.

{Abstract}: Ultrasonic technology was applied to extract mycelial polysaccharides from Paecilomyces hepiali and the process was optimized by response surface methodology. Antioxidant activity of polysaccharides was also investigated by 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity (%DPPHsc). Three independent variables were ratio of water to raw material (x(1)), ultrasonic power (x(2)) and extraction time (x(3)). Statistical analysis indicated that x(1), x(2), x(1)(2), x(3)(2), x(1)x(2), x(1)x(3) and x(2)x(3) had significant effect on the yields. Besides, x(2), x(1)(2) and x(2)(2) shaped the %DPPHsc of polysaccharides significantly. Three dimensional surface plots and contour plots were drawn from the mathematical model. The optimal conditions for yield and %DPPHsc were as follows: condition (1) x(1) = 125 mL/g, x(2) = 300 W, x(3) = 12 min, and condition (2) x(1) = 125 mL/g and x(2) = 500 W, x(3) = 11 min, respectively. Under these conditions, the yield was 9.37%, and %DPPHsc was 45.34%. There existed good agreement between experimental and predicted values.

{Abstract}: In this study, ethanol extracts of an edible mushroom Agaricus bisporus, before and after boiling were investigated for antioxidant, anticancer and antimicrobial activities. To confirm the total antioxidant activity, ABTS, DPPH free radical-scavenging assay was carried, along with total phenolic and flavonoid concentration. In measuring ABTS and DPPH free radical scavenging activities, scavenging activity was found to be similar in both the extracts. Total flavonoids were 16.4 +/- 0.5 and 15.2 +/- 0.2 mg/g (Quercetin equivalent), the phenolics were 90.2 +/- 0.6 and 70.6 +/- 0.1 mg/g (Catechin equivalent) in raw and boiled extract, respectively. A. bisporus extracts inhibited cell proliferation of HL-60 leukemia by the induction of apoptosis. In addition, A. bisporus extracts exhibited antibacterial activity against both gram positive and gram negative bacteria, as well as anticandidal activity against Candida albicans. Therefore, A. bisporus could be considered as a functional food with antimicrobial, anti-oxidative and antiproliferative activity.
